Hopefully the two of you (brothers) can talk frankly with each other to start. As Mark pointed out it would good to start by observing to your brother that the wife-to-be seems to be distant. It's kind of hard for us here to put our finger on it as we do not know the other people at all personally. Don't jump to conclusions too soon.
