Seems like it to me. If the differential only has to make up the difference in a turn it probably doesn"t get much use and maybe gets crudded up. Running it continuously and probably at a higher speed may cause this crud to make noise.
But, thinking more about it, the thing that contacts the road is the tread and the tread didn"t change it"s length (circumference), well maybe a little bit as the flat tire could become concave rather than be convex as it is in normal operation meaning the tread actually went up into the tire and it essentially ran on the sidewall. That could do it as the circumference would now be shorter than it was.
